Principle of Gel Permeation Chromatography and Steps in Gel Permeation Chromatography

The size exclusion technique of gel permeation chromatography is used to separate and characterise polymers. It is a one-of-a-kind technique for determining the molecular weight distribution of an analyte using polymer characterisation. The polymer's profile is significant because it affects physical qualities including brittleness, elasticity, and toughness in its final application. The premise behind the gel permeation chromatography system is that various components have variable affinity for the stationary phase. Smaller components become caught in the pores of the packing material, resulting in a long retention duration. Larger components in analytes, on the other hand, pass through the stationary phase without significant affinity, resulting in a shorter retention duration. Based on this information, the processing unit displays various peaks on the display that can be used for cauterization and separation of the analyte. Pump, injector, columns, detectors, and data processing systems are among the components of a gel permeation chromatography system.

Principle of Gel Permeation Chromatography

It's a technique for separating components based on their molecular weight or size differences.

The stationary phase is a porous polymer matrix with pores that are entirely filled with the mobile phase solvent.

The molecules in the sample are pushed through microporous packing material in specialised columns (gel).

The separation is based on the fact that molecules larger than a particular size are completely prohibited from the pores, while smaller molecules have partial or complete access to the interior of the pores.

As a result of the flow of the mobile phase, bigger molecules will travel down the column unobstructed, without piercing the gel matrix, but smaller molecules would be slowed based on their gel penetration.

Steps in Gel Permeation Chromatography

It consists of three major steps:

A. Column preparation for gel filtration

It entails:

The gel has swelled.

Semi-permeable, porous polymer gel beads with a well-defined range of pore diameters are packed into the column.

Washing: After packing, the column is washed with several column volumes of buffer solution to remove any air bubbles and test column homogeneity.

B. Using a syringe, load the sample onto the column.

C. Component detection and eluding the sample
